Default Joining rooms and adding value or not?

If by box room you mean walk-in cupboard (no windows, and thus not an
inhabitable room) then you will be bringing this into use as habitable
space, so it is likely to add value, especially if the medium room is on the
smaller side of medium.

However if the medium room is on the larger side of medium, you might be
better not losing a room (even an uninhabitable one) but thinking of eg
en-suite bathroom.

You will need building regulations approval for the removal of the wall and
to ensure that eg windows and ventilation are sufficient for the larger
habitable area created.
